EVs in a New Light: Single-EV phenotyping and cellular uptake at 20 nm resolution with ONI super-resolution microscopy
概要
Extracellular vesicles (EVs) are heterogeneous at the single-particle level, but bulk measurements can obscure differences in marker expression, size, and cellular behavior. This seminar will introduce compact super-resolution microscopy workflows for characterizing individual EVs and studying their interactions with target cells. Using the ONI Aplo Scope and Nanoimager, multicolor dSTORM can resolve and phenotype EV surface markers at 20 nm resolution. TIRF, HiLo, and widefield imaging can show EV localization in live or fixed cells, while single-particle tracking can quantify movement in live cells. Examples will show how these methods can support EV detection, phenotyping, colocalization, uptake studies, and quantitative comparison of heterogeneous populations. Practical considerations for fluorophore selection, sample preparation, imaging conditions, and data interpretation will also be discussed.
